Differences best heard with headphones or through decent speakers.

Same riff played four times using a parts strat with a JBJr humbucker in the bridge position
Riff 1: Tweed, Thin
Riff 2: Tweed, Fat
Riff 3: Tude, Thin
Riff 4: Tude, Fat

WGS ET65, TAD 6L6GC-STR, Tung Sol 12AX7 (new production)
Volume: 11
Bass: 6
Treble: 10
MV: 10
Level: 6

SM57 and e609, Presonus USB, Garageband. no effects or EQ. Dry signal.

Same riff played 5 times. Neck = Alnico Pro II, Middle = Duckbucker, Bridge = JBJr
Riff 1: Neck, Tweed, Thin
Riff 2: Middle, Tweed, Thin
Riff 3: Bridge, Tweed, Thin
Riff 4: Bridge, Tweed, Fat
Riff 5: Bridge, Tude, Fat

Same settings as above.
The pickup output clearly makes a difference on the amount of overdrive tones in Tweed mode. With traditional single coil, there's just a bit of "edge" in Tweed/Thin when the amp is cranked. However, the JBJr really takes it to the next level.
